“We introduce a novel dynamic model of stem cell and tissue organisation in murine intestinal crypts. Integrating the molecular, cellular and tissue level of description, this model links a broad

spectrum of experimental observations encompassing spatially confined cell proliferation, directed cell migration, multiple cell lineage decisions and clonal competition. Using computational simulations we demonstrate that the model is capable of quantitatively describing and predicting the dynamic behaviour of the intestinal tissue during steady state as well as after cell damage and following selective gain or loss of gene function manipulations affecting Wnt- and Notch-signalling. Etomoxir Our simulation results suggest that reversibility and flexibility of cellular decisions are key elements of robust tissue organisation of the intestine. We predict that the tissue should be able to fully recover after complete elimination of cellular subpopulations including subpopulations deemed to be functional stem cells. This challenges current views of tissue stem cell organisation.”
“Background selleck chemicals and Aims: Recent

evidence suggests that hyperuricemia might increase the risk of cardiovascular disease. Cardiovascular risk factors are well recognized to be associated with cerebral white matter lesion (WML). We hypothesized that hyperuricemia is related to higher grade of WML in both men and women.

Methods and Results: A total of 108 men and 123 women were enrolled from those who signed up for the annual Health Examination for the Elderly Program in Taipei from 2006 to 2008. Information in interview data, clinical and laboratory examinations were collected. Hyperuricemia was defined by uric acid

>= 458.0 mu mol/L in men and >= 392.6 mu mol/L in women. Two types of WML including periventricular hyper-intensity (PVH) and deep white matter hyper-intensity (DWMH), ascertained by cranial magnetic resonance imaging, were graded. Association between hyperuricemia and high grade WML was evaluated by using multiple logistic regression analysis.

The prevalences of hyperuricemia BB-94 clinical trial were 13.9% and 17.9% for men and women, respectively. The prevalences of moderate-to-severe PVH among men and women were 16.7% and 7.3%; while the prevalences of moderate-to-severe DWMH for men and women were 19.4% and 11.4%, respectively. Hyperuricemia was related to moderate-to-severe DWMH in men, after controlling for age, education years, smoking, alcohol consumption, metabolic Z score, silent infarct, and the use of anti-hypertensives, lipid-lowering and anti-diabetic agents, the association remained significant. The relationship was not evident among women.

“The effects of microbial transglutaminase (MTG) on heat-induced gelation of pork myofibrillar proteins (PMP) structural changes, textural properties were studied by Raman

spectroscopy and texture profile analysis (TPA), respectively. And the relationships between the structural changes and textural characteristics were estimated by principal component analysis (PCA). Changes in the Raman spectra were interpreted as the occurrence of secondary structural changes in myofibrillar proteins with MTG added. Modifications in the amide I (1600-1700 cm(-1)) regions indicated a significant (p<0.05) decrease in alpha-helix content, accompanied by a significant (p<0.05) increase in beta-sheets, beta-turns and random coil content due to the addition of

the enzyme. Obvious texture property changes were also determined by TPA. All these changes showed GSK2126458 a strong, irreversible heat-induced gel formed due to the addition of MTG. The application of a dimensionality reducing technique such as PCA proved to be useful to determine the most influential properties of heat-induced gel. Significant (p<0.05) correlations were found between these structural changes and the textural characteristics (hardness) in PMP system with the addition of MTG by PCA. The hardness was related positively to fraction of beta-sheet, beta-turns and random coil, and negatively to normalized intensity of 760 cm(-1) and fraction of a-helix. The samples are closely grouped in a cluster defined by level of MTG. (C) 2011 Elsevier Ltd. All rights reserved.”

“Background and Aim: The aim of the present work was to evaluate the relationship between energy-generating nutrients and the presence of central and overall obesity after correcting for socio-demographic, lifestyle and clinical characteristics, among healthy elders. Methods and Results: During 2005e2007, 553 elderly men and 637 elderly women (mean age 74 +/- 7 years) from eight Mediterranean islands in Greece and Cyprus, were enrolled. The retrieved information included demographic, bio-clinical and dietary characteristics. MedDiet-Score assessed adherence SB273005 mouse to the Mediterranean dietary pattern.

The prevalence of obesity was 27% in males and 39% in females (p < 0.001), while 73% of males and 87% of females had central

obesity. The prevalence of diabetes, hypercholesterolemia and hypertension was higher in the obese than in the non-obese participants (p < 0.01). After adjusting for various confounders, a 1% increase in carbohydrate consumption was associated with a 12% (95% CI 0.78-0.99) lower likelihood of having central obesity, while a 1% increase in carbohydrate

and protein consumption was associated with a 14% (95% CI 0.78-0.95) and 16% (95% CI 0.72-0.97) lower likelihood of being obese, respectively. Vegetable protein was found to be associated U0126 concentration with a 15% (95% CI 0.77-0.93) lower likelihood of being obese while, only low glycemic index carbohydrates seem to be associated with a 6% (95% CI 0.90-0.98) lower likelihood of having central obesity.

Conclusions: The presented findings suggest that a diet high in carbohydrates and vegetable protein is associated with a lower likelihood of being obese and may help elderly people to preserve normal weight. (C) 2009 Elsevier B.V. All rights reserved.”
“With Fe compensation, the heavy rare earth element Gd-filled Gd(y)Fe(x)Co(4-x)Sb(12) (x < 2) skutterudites have been successfully synthesized by melting-annealing approach. Fe substitution on the Co site brings two contrary effects on Gd filling: charge compensation which enhances the filling fraction of Gd, and Lattice expansion which is deleterious for the stability of filled compounds that contain smaller atoms. When Fe content is less than 1.7, pure Gd(y)Fe(x)Co(4-x)Sb(12) compounds are obtained and the Gd maximum filling fraction (y(max)) increases with Fe content. The power factor (S(2)sigma) of the Gd(y)Fe(x)Co(4-x)Sb(12) increases with Fe content.

“The reactions between terpenes and ozone (or other oxidants) produce a wide variety of both gas- and particle-phase products. Terpenes are biogenic volatile organic compounds (VOCs) that are also contained in many consumer products. Ozone is present indoors since it infiltrates into CX-6258 supplier the indoor environment and is emitted by some office and consumer equipment. Some of the gaseous products formed are irritating to biological tissues, while the condensed-phase products have received attention due to their contribution to ambient fine particulate matter (PM2.5) and its respective health

significance. Despite common scientific questions, the indoor and ambient air research communities have tended to operate in LY2157299 isolation regarding this topic. This review critically evaluates the literature related to terpene oxidation products and attempts to synthesize results of indoor and ambient air studies to better understand the health significance of these materials and identify knowledge gaps. The review documents the results of a literature search covering terpene oxidation chemistry, epidemiological, toxicological, and controlled human exposure studies, as well as health studies focused more generically on secondary organic aerosol (SOA). The literature shows a clear role for gas-phase terpene oxidation products in adverse airway effects at high concentrations; however, whether these effects occur at more environmentally relevant levels is unclear. The evidence for toxicity of particle-phase products is less conclusive.

Despite 5 tag SNPs were required to capture all variability, just one SNP allowed

to define both haplogroups, and only two SNPs were needed to differentiate the most common haplotypes. An additional taq SNP was necessary to identify both URR variants. Allele-frequency analysis of a selected taq SNP among breeds showed a geographical cline. European Bos taurus breeds had lower frequencies of the C allele than B. indicus type cattle, while Creole cattle and Wagyu breeds had intermediate frequency. There was a significant correlation between frequency profile and timing of puberty among the studied breeds, which seems to suggest that genetic variation within bovine GNRHR gene could explain at least INCB024360 clinical trial part of the reported variability. “Imide units are incorporated into thermoplastic and solvent-based polyurethane (PU) chains

to improve the thermal stability of PU. However, these poly(urethane-imide) (PUI) materials have poor processablity and suffer from solvent emission. To prepare easily processable and environmentally friendly PUI products, some waterborne PUIs are synthesized using a prepolymer process. A series of PUI dispersions will, 25 wt % solid content, viscosities of 7.5-11.5 cps, and particle sizes of 63-207 nm was prepared. The composition-property relationship of PUIs, including the solubility behavior of PUI cast films, and their thermal and mechanical properties were established. The solvent resistance and tensile strength of PUI film increased with the number of imide groups. All PUIs exhibited improved thermal stability but not char yield as the temperature increased. The inclusion of a little imide increased the decomposition temperature of PUI while maintaining the elasticity of the polymer, revealing successful translation of PUI into the water-based form. “Polydimethylsiloxane/polystyrene (PDMS/PS) blends were prepared by radical copolymerization of styrene (St) and divinylbenzene BVD-523 cost (DVB) in the presence of alpha,omega-dihydroxy-polydimethylsiloxane (PDMS), using

benzoyl peroxide as initiator. The PDMS/PS blends obtained by this method are a series of stable, white gums, when the feed ratio of PDMS to St is 60/40 and DVB to St is not more than 2.0 wt %. Elastomers based on PDMS/PS blends were formed by crosslinking PDMS with methyl-triethoxysilicane (MTES). The MTES dosage was much larger than the amount necessary for end-linking hydroxy-terminated chains of PDMS, with the excess

being hydrolyzed to crosslinked networks, which were similar to SiO(2) and acted as filler. Mechanical property measurements show that the elastomers thus formed exhibit Superior mechanical properties with respect to pure PDMS elastomer and the elastomers based on PDMS/PS system we prepared before. Moreover, investigations were carried out on the elastomers by extraction measurement and selleck products scanning electron microscopy (SEM). The extraction data show that the sol-fraction decreases with increasing the feed ratio of DVB to St. SEM observation demonstrates that the elastomer has a microphase-separated structure consisting of dispersed PS domains within a continuous PDMS matrix, and the extracted material exhibits a porous structure. “A high-performance of uniaxial alignment of electrospun nanofibers was realized learn more by introducing an inclined gap into dual collectors that consisted of two conductive strips. Because the two strips that were configured horizontally

and vertically had a height difference from the inclined gap, the electrospun nanofibers were sequentially suspended across the edges of strips in a well-aligned and regularly distributed form. Some parameters, such as concentration of solution, applied voltage, and spinning distance were considered for the successful suspension and formation of the aligned electrospun fibers. The method could improve the properties of nanofiber alignment and allow for easy transfer onto other solid substrates or devices. The alignment technique used polycaprolactone, which resulted in continuous and well-aligned nanofibers with diameters ranging from 500 to 700 nm. Furthermore, it is suggested that repetitive transfer be used to achieve a higher density of aligned nanofiber arrays. This would enlarge the applicability of nanofibers, especially for the tissue

“Background: Adhesion of keratinocytes is crucial for maintaining the integrity of the skin, as demonstrated by the number of dermatological disorders of genetic origin that are associated with a defect of basal keratinocyte adhesion. Integrins are the main component of the molecular networks involved in this phenomenon, but there are many www.selleckchem.com/products/azd1390.html others. In a recent description of proteins associated to caveolae at the plasma membrane of human basal epidermal cells, we demonstrated that GS-7977 datasheet CD98hc is localized with beta 1 integrin.

Objectives: We investigated the CD98hc proteins interactions and the role of CD98hc in keratinocyte adhesion.

Methods: CD98hc protein interaction was identified following co-immunoprecipitation and proteomic analysis using LTQ-FT mass spectrometer. Extinction of CD98hc gene expression using specific short hairpin RNA or over-expression of CD98hc lacking the beta 1 integrin binding site was used to evaluate

the role of this protein in keratinocyte fate.

Results: We show that CD98hc forms molecular complexes

with beta 1 and beta 4 integrins in primary human keratinocytes and, using immunofluorescence, that these complexes are Evofosfamide molecular weight localized at the plasma membrane, in keeping with a role in adhesion. We confirmed that this protein is a key player of keratinocyte adhesion because in absence of interaction between CD98hc and integrins, beta 1 integrin failed to translocate from the cytoplasm to the plasma membrane and keratinocytes expressed epidermal differentiation markers.

Conclusions: All these data strongly suggested that CD98hc is involved in integrin trafficking and by consequence, in keratinocyte adhesion and differentiation. (C) 2011 Japanese Society for Investigative Dermatology. Published by Elsevier Ireland Ltd. All rights reserved.”

“P>The Tic20 protein was identified in pea (Pisum sativum) as a component of the chloroplast protein import apparatus. In Arabidopsis, there are four Tic20 homologues,

termed atTic20-I, atTic20-IV, atTic20-II and atTic20-V, all with predicted topological similarity to the pea protein (psTic20). Analysis of Tic20 sequences from many species indicated that they are phylogenetically unrelated to mitochondrial Rabusertib Tim17-22-23 proteins, and that they form two evolutionarily

conserved subgroups [characterized by psTic20/atTic20-I/IV (Group 1) and atTic20-II/V (Group 2)]. Like psTic20, all four Arabidopsis proteins have a predicted transit peptide consistent with targeting to the inner envelope. Envelope localization of each one was confirmed by analysis of YFP fusions. RT-PCR and microarray data revealed that the four genes are expressed throughout development. To assess the functional significance of the genes, T-DNA mutants were identified. Homozygous tic20-I plants had an albino phenotype that correlated with abnormal chloroplast development and reduced levels of chloroplast proteins. However, knockouts for the other three genes were indistinguishable from the wild type. To test for redundancy, double and triple mutants were studied; apart from those involving tic20-I, none was distinguishable from the wild type. The tic20-I tic20-II and tic20-I tic20-V double mutants were albino, like the corresponding tic20-I parent. In contrast, tic20-I tic20-IV double homozygotes could not be identified, due to gametophytic and embryonic lethality. Redundancy between atTic20-I and atTic20-IV was confirmed by complementation analysis.

Telephone-based, individual smoking cessation counseling has been shown to result in successful quit attempts in the general population Nepicastat mw and among pregnant women, but “”quitlines”" are underutilized.

A social marketing campaign called One Tiny Reason to Quit (OTRTQ) promoted calling a quitline (1-800-QUIT-NOW) to pregnant, African American women in Richmond, Virginia, in 2009 and was replicated there 2 years later.

Methods: The campaign disseminated messages via radio, interior bus ads, posters, newspaper ads, and billboards. Trained volunteers also delivered messages face-to-face and distributed branded give-away reminder items. The number of calls made from pregnant women in the Richmond area during summer 2009 was contrasted with (a) the number of calls during the seasons immediately before and after the campaign, and (b) the number of calls the previous summer. The replication used the same evaluation design.

Results:

There were statistically significant spikes in calls from pregnant women during both campaign waves for both types of contrasts. A higher proportion of the calls from pregnant women were from African Americans during the campaign.

Conclusion: A multimodal quitline promotion like OTRTQ should be considered for geographic areas with sizable African American populations see more and high rates of infant mortality.”
“Background: Fractures of the lateral clavicle result in a high rate of nonunion compared with midshaft fractures. Many operative treatment methods of unstable lateral clavicle fractures have been described, but no single method has become generally accepted. This study reports our experience treating unstable lateral clavicle fractures with coracoclavicular ligament rupture.

Methods: Within 7 years, 19 patients with acute unstable selleck kinase inhibitor lateral clavicle fractures were treated using a 2.4-mm T-plate and a coracoclavicular polydioxanone cord. The follow-up averaged 5.3 years

(range, 2.0-8.8). Shoulder function was assessed using the Constant score and Disabilities of the Arm, Shoulder and Hand score. General health of the patients was evaluated according to the Short Form 36 questionnaire.

Results: Eighteen of the 19 patients treated with our method achieved union at 8.8 weeks (range, 6-14), a Constant Murley score of 91.5 (range, 72-100) on the injured side versus 93.5 (range, 80-100) on the contralateral side, a Disabilities of the Arm, Shoulder and Hand score of 1.4 (range, 0-9.2), and a Short Form 36 Health Score of 85 (range, 89-100). One patient showed breakage of the plate and a delayed union at the radiologic follow-up 4 months after surgery.

Conclusion: This study shows that treatment of unstable lateral clavicular fractures with an angular stable locking plate together with a polydioxanone cord ligament augmentation leads to excellent results with a 95% union rate.
